Applications are invited from suitably qualified persons for the
position of Postdoctoral Fellow in the Departments of Zoology and
Biochemistry, University of Otago, Dunedin, New Zealand. This
position in the laboratories of Dr David A. Wharton (Zoology) and Dr.
Craig Marshall (Biochemistry), is funded by the Marsden Fund and is
available for three years. Associate investigators on the project
are Dr. Hans Raml=F8v, Roskilde University, Denmark and Prof. John
Barrett of the University of Wales.

The research project entitled "Ice active compounds and the survival
of intracellular freezing in an Antarctic nematode" will involve the
isolation and sequencing of a protein which is involved in the
ability of an Antarctic nematode to survive intracellular freezing
and the investigation of its properties and evolution. The appointee
will have experience in protein purification and molecular biology
techniques. We wish to fill this position by February 2001.

Applicants must have been awarded the degree of Doctor of Philosophy
before taking up this position. The salary for Postdoctoral Fellows
is NZ$46,350 per annum (rates effective 1 February 2001).

To Whom It May Concern,

My name is Jim Campbell, and I am working on my MS in Biology at Angelo
State University in San Angelo, Texas. The project focuses on the
phylogeny
of collared lizards and their parasites. One of the parasites is an
endoparasitic worm. I hope to amplify segments of its mt genome, but I
have
had difficulty performing successful genomic extractions. I have
isolated
the DNA with a protocol from Blouin, and the DNA is of the best quality
of
any extractions I have ever seen. This process includes only lysis
buffer
and Proteinase K treatment. Supposedly, the lysate can be used directly in
a
PCR. First of all, the DNA degrades several days after incubation with
Proteinase K, and DNA concentrations are initially low. Secondly, I have
been using degenerate primers that supposedly amplify regions of mtDNA
from
many animals, but the PCR refuses to amplify anything. Not even smudges
appear on agarose gels. I would greatly appreciate any guidance with my
genomic preps - a protocol or citation of a protocol. In addition, any
PCR
primer sequences that you could give me might cure the amplification
problem.

1) does your PCR reaction actually work? Positive control?.

2) degenerate primers?....Ta=25 C and you should have lots of bands.

Dr. Siddiqi,

We have two copies of this collection in our department. I looked at
them
and as far as I can tell these were individual publications published as
a
series over a number of years. I think that the whole series was later
bound together for convenience by individual libraries or departments,
similar to what is often done with journals from a particular year. It
is
my feeling that they should be cited as individual publications and not
as
a book.

I am studying ectoparasitic nematodes (Noctuidonema, Vampyronema,
and Acugutturus) of moths and cockroaches. A friend in the UK is
planning a trip to Belize to collect moths and he has agreed to collect
some for me to examine for nematodes. However, the usual preservative,
formalin, is toxic and there are restrictions on shipping it. We both
doubt
that a box of vials containing formalin would pass customs. He is not
certain whether he would be able to find any or (if he did) to prepare it
as a
10% solution buffered to pH=7. An alternative is alcohol (ethanol,
methanol, or iso-propanol). Have any of you had experience in
preserving
nematodes in alcohol for eventual mounting and microscopic
examination/photography? All my specimens have been preserved in either
buffered formalin or TAF and I have no experience with examination of
alcohol-preserved material. Any insights or references to procedures
involving nematodes killed in alcohol will be appreciated, as will
suggestions for shipping formalin to Belize. Additionally, I will be
interested
in any information or samples of ectoparasitic nematodes of moths and/or
cockroaches, particularly from Africa and Asia.
